摘要
取不同基因型的玉米自交系芽尖接种在MS(大量 ) +B5(微量 ) +6 BA(0 .1~ 2mg·L-1) +2 ,4 D(0 .5~4mg·L-1) +IAA (1mg·L-1)的诱导培养基上 ,结果发现玉米芽尖愈伤组织及丛生芽的形成能力与基因型及激素配比有关。通过RAPD分析 ,发现正常再生植株与畸形苗和白化苗的基因组之间存在差异 ,说明在愈伤组织诱导及继代过程中玉米的基因组发生了变异。通过扫描电镜观察发现玉米芽尖诱导再生植株有两种情况 :一是通过器官发生途径再生绿苗 ;二是经过体胚发生途径再生绿苗。
The shoot tip of different genotypes of inbreds of Zea mays L.were cultured in the induction media, which consists of MS(macroelements), B 5(microelements), 6 BA (0.1~2 mg·L -1 ), 2,4 D(0.5~4 mg·L -1 ) and IAA(1 mg·L -1 ). The generation of calli and multiple shoot clumps showed significant difference in various maize genotypes and hormone combinations. The results of randomly amplified polymorphic DNA(RAPD) showed that the genomic DNA was modified in tissue culture. Two patterns of generation of maize were found by observation with scanning electron microscope. One was induced from organogenesis and the other produced somatic embryogenesis after subculturing several times.
