摘要 目的 :分析 1株对亚胺培南耐药的铜绿假单胞菌的耐药机制。方法 :用改良的三相水解试验分析该菌株所产的 β内酰胺酶 ,并进行初步分类 ;用聚合酶链反应 (PCR)扩增该菌的耐药基因 ,用PCR 限制片段长度多态性 (RFLP)初筛整合子并分型 ,用长片段PCR扩增整合子的可变区并测序 ,以确认 β内酰胺酶基因种类及其转移特性。 结果 :发现铜绿假单胞菌RJ2 1 3携带一个整合子介导的blaVIM 2基因。结论 :在上海和中国大陆地区首次发现铜绿假单胞菌产生的整合子介导VIM 2型金属 β内酰胺酶 。 Objective: To analyze the resistant mechanism to imipenem in a strain of Pseudomonas aeruginosa . Methods:Using modified three dimension test to analyze and classify the β lactamase produced by the strain; detecting the resistance genes by PCR; to screen and classify the integron by PCR RFLP; to amplify the variable region of integron by long fragment PCR; the PCR products were sequenced subsequently. Results:Clinical strain of Pseudomonas aeruginosa RJ213 carrying an integron mediated vim 2 β lactamase was found. Conclusions:Integron mediated VIM 2 type metallo β lactamase exists in Shanghai, clinical isolate which cause resistance to imipenem.
