摘要
为了探讨增骨Ⅰ 号的作用机制 ,进行了体外成纤维细胞的实验。采用酶消化法将人包皮组织进行成纤维细胞的分离 ,纯化及培养 ,并测定骨钙素、细胞钙含量。结果显示成纤维细胞在增骨Ⅰ 号组增殖迅速 ,其OD值明显高于对照组 (P <0 .0 5 ) ,但反映成骨表型表达的骨钙素及细胞钙含量未见明显增加。表明增骨 Ⅰ 号仅有激活成纤维细胞的作用 ,本实验结果为序贯疗法应用增骨 Ⅰ 号作为骨重建中的激活药物提供了理论依据。
In order to explore the action mechanism of 'ZENG GU' No. 1 (ZG No. 1, a TCD preparation), fibroblasts (FBS) were experimented in vitro: isolated, purified and cultivated from human foreskin with trypsinase digestion method; and the osteocalcin (BGP) and calcium content in FBS were determined. The results showed that the FBS proliferation in ZG No. 1 group was rapid with OD value obviously higher than that in the control group (P < 0.05) but no significant increase was found in the BGP and calcium content reflecting the osteogenic phenotypic expression, suggesting that ZG No. 1 functions only to activate FBS; the results of this study could provide a theoretic basis on which ZG No. 1 may be used as the activating drug for bone remoulding in the sequential therapy.
