摘要
目的:探讨肠复康对人结肠癌HT29裸小鼠移植瘤基质金属蛋白酶-2(MMP-2)、金属蛋白酶组织抑制因子-2(TIMP-2)的影响,以此对肿瘤康复领域的功能性改良措施的选择提供有利依据。方法:建立人结肠癌HT29癌细胞株裸小鼠皮下移植瘤模型,采用随机数字表法随机分成3组,模型组8只,肠复康组7只,西药组8只。免疫组化染色结合图像分析系统半定量检测移植瘤组织内MMP-2、TIMP-2积分光密度(IOD)及MMP-2/TIMP-2比值。结果:与模型组比,肠复康组可使移植瘤MMP-2和TIMP-2含量增高,前者无显著差异(P>0.05),后者具明显差异(P<0.01),西药组也能增高移植瘤MMP-2和TIMP-2含量,但皆无明显差异(P>0.05);肠复康组和西药组可降低MMP-2/TIMP-2比值,前者有显著差异(P<0.05),后者无明显差异(P>0.05)。结论:肠复康可降低移植瘤内MMP-2/TIMP-2比值,对人结肠癌HT29裸小鼠移植瘤细胞外基质(ECM)降解可能具有抑制作用。
AIM:To investigate effect of Ch angfukang on matrix metalloproteinase-2(MMP-2)and tissue inhibitors of met allopro teinase-2(TIMP-2)of trans-planted tumor in the nude mice by huma n colon cancer HT29cells so as to provide favorable evidence for the s electio n of functional improvement in the field of tumor rehabilitation.METHODS:Aftr the model of the transplanted t umor of subcutaneous tis-sues located the nud e mice was establ ished by human colon cancer HT29cells,HT29nude mice were randomly divided into three groups:model group of 8mice,Changfukang group of 7mice and western medicine g roup of 8mice.The integra optical density(IOD) of MMP-2and TIMP-2in the tumor tissu es and the ratio of the MMP-2to the T IMP-2we re detected semi-quantitatively by immunohistochemical staining comb ined with image analysis.RESULTS:Compared with the model group,the co ntents of MMP-2and TIMP-2in the tumour tissues were increased in the Changfukang gr oup(between the two groups,no significant difference of MMP-2,P>0.05; significant difference of TIMP-2,P<0.01),and were also increased in th e western medicine group(between the two groups,no significa nt difference of both MMP-2and TIMP-2,P>0.05).The ratio of MMP-2to TIMP-2in the t umour tissues was increased in the Changfukang group and the western medicine gr oup(significant difference in the forme r group,P<0.05;no significant di fference in the latte r group,P>0.05).Changfukang can reduce the ratio of the MMP-2to th e TIMP-2in transplanted tumor tissues and may i nhibit decomposition of th e extracelluar matrix(ECM)of transplanted tumor tis sues in the nude mice by human colon cancer HT29cells.
