三维培养的人成纤维细胞增殖特性在皮肤组织工程研究中的意义

Significance of proliferation of human dermal fibroblasts after three-dimensional culture in the skin tissue engineering research

目的:探讨三维培养对人皮肤成纤维细胞增殖的影响。方法:培养人皮肤成纤维细胞,制备鼠尾胶原。对人皮肤成纤维细胞进行三维培养,检测细胞的增殖特性。结果:三维培养的人皮肤成纤维细胞生长曲线显示细胞增殖缓慢,流式细胞仪检测7,14dG0/G1期细胞分别为(79±3)%,(87±2)%。未见明显细胞凋亡峰。结论:三维培养对人皮肤成纤维细胞增殖有抑制作用。

AIM:To investigate the effect of three-d imensional culture on th e prolifera-tion of human dermal fibroblasts.METHODS:Human dermal fibroblasts w ere cultu red and the rat tail colla-gen were prepared.The proliferatio n char acteristics of human dermal fibrob-lasts were detected after three-dim ensiona l culture.RESULTS:The proliferation of fibroblasts wa s slow,observed through the grow curve of human dermal fibroblasts after three-dimensional cul-ture. Detected by flow cytometer(FCM),the cells in G 0 /G 1 stage were(79±3)%and(87±2)%at 7and 14day respectively and n o ap optosis peak was observed.Three-dimensional culture can inhi bit the proliferation of human dermal fibroblasts.
