摘要
目的:观察紫外线(Ultraviolet,UV)对体外培养的角质形成细胞株(HaCaT细胞)细胞因子分泌的影响。方法:UV照射佛泊脂和脂多糖(PMA/LPS)刺激的HaCaT细胞,用ELISA法检测细胞培养上清液中细胞因子IL-1α,IL-2,IL-6和IL-8的含量。结果:正常HaCaT细胞有IL-1α,IL-2,IL-6和IL-8的自发性分泌。PMA和LPS刺激HaCaT细胞,其IL-1α分泌量下降(P<0.05),IL-6和IL-8分泌量明显增加(P<0.01)。正常HaCaT细胞经UV照射,IL-1α,IL-2,IL-6的分泌无明显变化(P>0.05),IL-8分泌量升高(P<0.05)。PMA和LPS刺激的HaCaT细胞经UV照射,其IL-1α,IL-6和IL-8的分泌量明显增加(P<0.01)。结论:UV对不同状态下HaCaT细胞的作用是不同的。
AIM:To observe the e ff ect of ultraviolet (UV)on cytokine secretion in human keratinocyte HaCaT cell line in both normal condition and after stimulation with PMA and LPS.METHODS:T he expression of IL-1α,IL-2,IL-6and IL-8was detected by enzyme-linke d immunosorbent assay(ELISA).RESULTS:Normal HaCaT cells secreted IL-1α,I L-2,IL-6and IL-8spon-taneously.Stimulation by PMA and LPS induced incre ase in IL-6and IL-8production(P<0.01)and decrease in IL-1αlevel(P <0.01)in HaCaT cells.UV exposure of normal HaCaT cells only increased the secretion of IL-8obviously(P<0.01),which may participate in the infla m mation in-duced by UV in normal skin.In HaCaT ce ll stimulated by PMA and LP S,UV exposure increased the secretion of IL-1α,IL-6,and IL-8.The effect of UV on normal and stimula ted HaCaT cell line can be different .
