长寿花组织培养与人工种子的研究 被引量:4

摘要 笔者初步研究了长寿花人工种子的制备,包括胚性愈伤组织及胚状体的诱导、人工种子的包埋和萌发。胚性愈伤组织诱导的最适培养基为:MS+2,4 D2mg/L+BA0 2mg/L+蔗糖30g/L+琼脂6g/L;胚状体诱导的最适培养基为:MS+BA2mg/L+NAA0 2mg/L+活性炭4g/L+蔗糖30g/L+琼脂6g/L;用无激素MS培养基配制的4%海藻酸钠溶液包埋胚状体,并用2%的CaCl2·2H2O溶液作聚胶化因子,制成人工种子;将人工种子在无激素的MS固体培养基上萌发,萌发率达96%。利用石蜡切片法对胚状体的发生发育过程进行了观察。 The study aimed at production of the Kalanchoe flammea′s artificial seed,including the selection of embryonic callus, the inducement of embryoid,embedding and sprouting of artificial seed . During inducing embryonic callus the optimum medium was MS + 2,4-D 2 mg/L + BA 0.2 mg/L. During the inducting of embryoid, the culture mediums with different regulator concentrations were used, finally the culture medium with the highest of inducement frequency was MS + BA 2 mg/L + NAA 0.2 mg/L+ active carbon 4 g/L. In making artificial seed, sodium alginate in 4% concentration was used as embedding agent, and CaCl2·2H2O in 2% consistency as gel assembled factor. The sprouting rate of artificial seed on MS without regulator is up to 96%. The appearance and growing of embryoid were observed through paraffin slices of callus.
