摘要
采用超声波法、索氏法、改良索氏法3种方法分别从金银花中提取绿原酸,用紫外光谱法和高效液相色谱法对其进行定性定量分析。同时用金银花乙醇提取物及纯化的绿原酸对乳房炎主要致病菌进行抑菌试验。提取试验结果表明:改良索氏法为金银花中绿原酸提取的最佳方法,获得绿原酸的提取率显著高于其他方法(P<0 01),提取终产物与标准品的最高吸收峰均在327nm处,终产物中绿原酸的含量为64 01%。抑菌试验结果表明:金黄色葡萄球菌对金银花醇提物极敏感,乳房链球菌、产气荚膜梭菌对其高敏,无乳链球菌、停乳链球菌对其中敏;绿原酸对金黄色葡萄球菌、无乳链球菌、停乳链球菌、乳房链球菌、产气荚膜梭菌的最小抑菌浓度(MIC)分别为1 58g/L、3 16g/L、3 16g/L、6 32g/L、1 58g/L,最小杀菌浓度(MBC)分别为1 58g/L、6 32g/L、3 16g/L、6 32g/L、3 16g/L。
Optimization of the extracting method of Chlorogenic Acid from Flos Lonicerae was studied by three different methods of ultrasonic extraction, sorbitic extraction, and improved method of sorbitic extraction. The quantative and qualitative determination of Chlorogenic acid in Flos Lonicerae has been carried out by two different ways of Ultraviolet and Visible Spectrophotometric(UV) and High Performance Liquid Chromatographic(HPLC). Inhibition test against pathogenic bacteria has been done with Flos Lonicerae ethanol legging and Chlorogenic acid. The results showed that Chlorogenic acid tiptop absorb-apex was 327 nm, and improved method of sorbitic extraction was superior remarkably to other methods (P<0.01), and the content of Chlorogenic acid of witch eventual producetion was disdilled by the improved method of sorbitic extraction was 64.01%. So the improved method of sorbitic extraction is the best way. The antibacterial action in vitro to pathogenic bacteria caused mastitis with Flos Lonicerae ethanol legging showed that Staphylococcus aureus was extremely sensitive, and Streptococcus uberis, Clostridium aerogenes capsulatus were highly sensitive, and Streptococcus agalactiae, Streptococcus dysgalactiae were moderately sensitive. The minimal inhibitory concentration (MIC) of Chlorogenic acid performed by the tube and plate method to Staphylococcus aureus, Streptococcus agalactiae, Streptococcus dysgalactiae, Streptococcus uberis, Clostridium aerogenes capsulatus were 1.58 g/L, 3.16 g/L, 3.16 g/L, 6.32 g/L, 1.58 g/L respectively and minimal bactericidal concentration (MBC) were 1.58 g/L, 6.32 g/L, 3.16 g/L, 6.32 g/L, 3.16 g/L
