活血方对免疫介导再生障碍性贫血小鼠IL-3、IFN-γ的影响 被引量:2

摘要 目的 检测免疫介导再生障碍性贫血(再障)小鼠骨髓造血细胞在含活血药血清液体培养体系中IL-3、IFN-γ的浓度,探讨活血方药治疗再障的作用机制。方法 用活血方中药煎剂灌喂SD大鼠,抽取、制备含中药血清;Balb/c小鼠经60Co 5.5 Gy γ射线照射后,由尾静脉输入取自DBA/2小鼠胸腺淋巴结混合细胞悬液,形成免疫介导再障模型。在含活血药血清的液体培养体系中培养再障小鼠骨髓造血细胞,检测培养体系中IL-3、IFN-γ的浓度。结果 正常活血血清组IL-3浓度显著高于正常对照血清组(P<0.01),而再障活血血清组IL-3浓度则显著低于再障对照血清组(P<0.01),正常活血血清组、正常对照血清组IFN-γ均为0。再障活血血清组、再障对照血清组两组间IFN-γ无差异(P>0.05)。结论 活血方药临床治疗再障的作用可能是影响IL-3的生成而进一步促进造血干/祖细胞增殖、分化。 Purpose: To study the mechanism of treatment of aplastic anemia with promote-blood-flow decoction. Methods: We raised SD rats with promote-blood-flow decoction to prepare the serum containing Chinese herbal materia. Radiated by 60Co 5.5 Gy γ ray, Balb/c mice were given mixed cells from thymus and lymph nodes of DBA/2 mice through tail vein to establish the immune - mediated aplastic mice model. The hemopoietic cells of aplastic mice were cultured in the liquid cell culture system containing promote-blood-flow decoction. Then we measured IL-3, IFN-γ in the different culture systems. Results: IL-3 was significantly higher in the normal promote-blood-flow serum group than in the normal control serum group (P0.05). Conclusions: Promote-blood-flow decoction maybe has effect on the production of IL-3 to compact on proliferation and differentiation of stem/progenitor cells.
