摘要
为进一步筛选、克隆和表达牦牛相关基因构建了牦牛肝脏的cDNA文库,从牦牛肝脏中提取、分离mRNA,逆转录合成cDNA后,以pSPORT1质粒为载体构建cDNA文库.经鉴定,牦牛肝脏cDNA 文库的库容量为1×106转化子/μg cDNA,以菌株平板法测定,重组率为93%.所构建的cDNA 文库容量符合建库要求,能够用于目的基因筛选.
Yak is a very useful animal. The cDNA library of yak liver was constructed,which made a preparation for further cloning and expressing of wanted genes. mRNA from yak liver was extracted and purified,and cDNA was obtained by reverse transcription,and then cDNA library was constructed using plasmid pSPORT1 as the vector. The volume of the library was 1 × 106 transformants/μg cDNA and the rate of recombination was 93%. The results indicated that the cDNA library had enough volume for selecting wanted genes and made the basic of studying yak s genetic background.
