摘要
目的 :观察不同浓度的二贝母胶囊对S180 、EMT6瘤株的体内、体外抑瘤作用 .方法 :体外抑瘤试验采用MTT法 ;体内试验采用小鼠移植瘤法 ,瘤株为鼠源性S180 肉瘤及EMT6乳腺癌细胞株 ,治疗采用小鼠ig 12d并比较合并 5GY60 Co γ射线治疗的效果 .体外抑瘤试验观察指标为不同浓度的二贝母胶囊对MTT还原产物吸光度值的影响 ;体内抑瘤试验观察指标为肿瘤移植前后的小鼠体质量、瘤重、脾重 ,合并60 Co γ射线治疗时亦观察外周血WBC数量 .结果 :MTT试验显示对于EMT6细胞株 ,二贝母胶囊在 6 .2 5~ 10 0 g·L-1浓度范围内对MTT还原产物吸光度有降低作用 (P <0 .0 1或P <0 .0 5 ) ,最佳抑制浓度为 2 5 g·L-1.ig剂量为 2g·kg-1的二贝母胶囊 ,给药 12d后 ,对荷S180 及荷EMT6瘤株小鼠的肿瘤生长抑制明显 ,给药组瘤质量为 (0 .98± 0 .4 9) g及 (0 .6 2± 0 .38) g ;而生理盐水组瘤质量为 (1.6 2± 0 .4 5 ) g及 (1.2 2± 0 .5 2 )g ,瘤质量抑制率分别达到 39.5 %及 4 9.2 % ,合并放疗对S180 瘤株瘤质量抑制率达 5 3.2 % ,且使外周血WBC及脾质量维持在正常水平 .结论 :二贝母胶囊有抗肿瘤及免疫增强作用 .
AIM: To observe the antineoplastic effect of Erbeimu capsule on S 180 , EMT 6 both in vitro and in vivo . METHODS: For in vitro test, MTT method was used to examine the effect of different concentration of Erbeimu capsule on OD value of the reduced products. For in vivo test, the effect of Erbeimu capsule on S 180 and EMT 6, the body weight, tumor weight, spleen weight were measured before and after ig Erbeimu capsule. When radiotherapy with 5 GY 60 Co γ irradiation was also given, WBC in peripheral blood was also examined. RESULTS: MTT showed that ErBeiMu capsule significantly inhibited function of EMT 6 cell clone at concentration of 6.25-100 g·L -1 ( P <0.01 or P <0.05), which reached the maximum at 25 g·L -1 . After ig Erbeimu at the dose of 2 g·kg -1 to mice with transplatation tumor for 12 d, the tumor weight of S 180 and EMT 6 was (0.98 ±0.49) g and (0.62±0.38) g, while that in normal saline group was (1.62±0.45) g and (1.22±0.52) g respectively, with the inhibition rate reaching 39.5% and 49.2% respectively. When combined with radiotherapy, the inhibition rate reached to 53.2%, and WBC and the weight of spleen were kept at the normal level. CONCLUSION: Erbeimu -capsule has anti-tumor and immune enhancement effect.
