摘要 目的 :了解雌激素与流体剪切力对体外培养的成骨细胞增殖和功能的间接影响 .方法 :进行新生SD大鼠颅盖骨成骨细胞原代培养 .选择恒定的雌激素浓度及合适的振荡频率分四种组合方式施加于成骨细胞 ,2 4h后用其培养液培养静止状态下的成骨细胞 ,观察不同时间段细胞增殖和碱性磷酸酶活性的变化 .结果 :经雌激素和机械力影响的条件培养液作用 12h后 ,可促进细胞增殖 (P <0 .0 1) ,该效果持续至 2 4h ,在 2 4h至 72h两因素表现为协同作用 (P <0 .0 1) ;对ALP的活性的影响在 12h之前两因素表现为拮抗作用及抑制作用 (P <0 .0 1) .2 4h后 ,机械力可促进ALP活性升高 ,并持续至 72h .结论 AIM: To observed the indirect effect of estrogen and/or wall shear stress on rat osteoblasts in vitro . METHODS: Isolated and purified osteoblasts from the calvaria of newborn SD rats were primarily cultured, and 3 to 4 passages were used. Estrogen and wall shear stress were used 24 h on four groups of osteoblasts:① estrogen and wall shear stressed cells; ② wall shear stressed cells; ③ estrogen cells; ④ control cells. The media from these four groups were collected and were used on other four groups of osteoblasts. The proliferation and ALP activity of the cells from 6 to 72 h were observed. RESULTS: The condition media from the estrogen cells and the wall shear stressed cells both increased proliferation of the cells from 12 h to 24 h. And the media from the combination of these two factors further increased the proliferation of the cells from 24 to 72 h ( P <0.01). ALP activity was increased by the medium of stress after 24 h and the two factors one decrease ALP activity in the early stage (from 6 h to 12 h). CONCLUSION: Estrogen and wall shear stress can influence the proliferation and activity of osteoblasts by the way of paracrine. Osteoblasts can secrete factors into the medium under the stimulation of wall shear stress.
