摘要
目的 :研究柯萨奇 B4 病毒感染对人胚胰岛的影响。方法 :采用中国流行的柯萨奇 B4 病毒感染体外培养的人胚胰岛细胞 ,电镜观察其形态变化 ,并进行糖刺激的胰岛素释放量试验。结果 :病毒可直接攻击胰岛 ,突破胰岛胞膜 ,侵犯胰岛β细胞 ,可在β细胞内繁殖 ,使β细胞脱颗粒。糖刺激的胰岛素释放量在感染后 2 4 h增加 ,以后呈逐渐降低趋势。感染组在感染 2 4 h时低糖及高糖刺激的胰岛素释放量分别为 (6 4 .5 0± 9.2 0 ) m U· L- 1 ,(87.85± 8.5 0 ) m U· L- 1 ,均高于对照组 (P<0 .0 5 )。 4 8h~ 6 d高糖及低糖刺激的胰岛素释放量均显著低于对照组 (P<0 .0 5或 P<0 .0 1 )。结论 :柯萨奇 B4 病毒可在体外感染人胚胰岛细胞 ,并损害胰岛β细胞合成胰岛素的功能。
Objective To study the effect of CB 4V infection on human embryonic islet cell. Methods Human embryonic islet cell was infected by CB 4V in vitro , the morphological change of that was observed by electro microscope, and insulin releasing test by sugar stimulation was also proceeded. Results CB 4V could attack islet directly ,break the envelope of islet, invade β cell and proliferated in β cell. The quantity of insulin releasing stimulated by sugar increased 24 h after infection, then decreased gradually. The quantities of insulin releasing stimulated by hypoglycemia and hyperglycemia in infected group were (64 50±9 20)mU·L -1 and (87 85±8 50) mU·L -1 ,respectively,24 h later after infection, which showed significant difference compared with the control group ( P <0 05). From 48 h to 6 d, the quantities of insulin releasing stimulated by hypoglycemia and hyperglycemia were much lower than that of control group ( P <0 05, P <0 01).Conclusion CB 4V can infect human embryonic islet cell in vitro , and damage the function of islet β cell of secreting insulin.
