摘要 目的:研究在中波紫外线(UVB)诱导正常永生化角质形成细胞(HaCaT)凋亡过程中,T细胞凋亡相关基因19(TFAR 19)的表达时相及位置。方法:使用流式细胞仪、膜联蛋白V(Annexin-V)、激光扫描共聚焦显微镜等方法进行研究。结果:在凋亡过程中,TFAR 19逐渐向核周聚积,最后向核膜内移位,在核膜及其周围以及核内表达。 结论:在UVB诱导HaCaT细胞凋亡的早期和晚期过程中TFAR 19均有表达。 Objective: To study the expression of a new apoptosis-related gene TFAR 19 in HaCaT cell induced by UVB. Methods: Flow cytometric analysis, AnnexinV, laser confocal microscope were used in the study. Results: Apoptosis of HaCaT cell was observed 24 h after UVB irradiation, the number of apoptotic cells increased with the increase of the dose of irradiation, the higher the dose of UVB was, the earlier the appearance of apoptosis. Along with the increase of doses of UVB, the amount of TFAR 19 protein increased. The expression of TFAR 19 was observed around the nuclei of HaCaT cells, on the nuclear membrane and the nuclei. Conclusions: Apoptosis of HaCaT cells is induced by UVB, and TFAR 19 is expressed both in the early and late, phase of apoptosis.
