摘要
目的 探讨外源性核酸对自然衰老小鼠胸腺细胞IL 7mRNA和CD12 7(IL 7受体 )表达的影响。方法 10月龄雌性Balb c小鼠按体质量随机分为高剂量组 (每日灌胃 333.33mg·kg- 1 ·d- 1 鲑鱼鱼白DNA)、低剂量组 (每日灌胃 16 6 .6 7mg·kg- 1 ·d- 1 鲑鱼鱼白DNA)和对照组 ( 0 .1mol L的柠檬酸钠 )。 5周后测定胸腺脏器指数 ;原位杂交法检测胸腺内IL 7mRNA的表达 ,并用Image proPlus专业图像分析系统 ( 4 .0版 )对其表达面积百分比进行分析 ;间接免疫荧光流式细胞术检测老龄小鼠胸腺细胞CD12 7的表达。结果 鲑鱼鱼白DNA提取物对小鼠的体质量、胸腺质量和胸腺指数均没有影响 ,但高剂量SMD补充组可显著增加胸腺细胞IL 7mRNA表达和CD12 7淋巴细胞的数量 (分别P <0 .0 5 )。结论 补充鲑鱼鱼白DNA可能促进IL 7的产生及其受体的表达 ,从而延缓Balb
Objective To study the effect of exogenous nucleic acid extract on the expression of thymocyte IL 7 mRNA and CD127(IL 7 receptor) in natural aging mice. Methods Female Balb/c mice aged 10 months were divided randomly into three groups according to their weights. They were high dosage group(supplied 333.33 mg·kg -1 ·d -1 Salmon milt DNA),low dosage group(supplyied 166.67 mg·kg -1 ·d -1 Salmon milt DNA) and control group(supplyied 0.1 mol/L sodium lemonade). After five weeks, their thymus indices were measured. The expression of IL 7mRNA was detected by in situ hybridization and percentage of expression area was analyzed using Image Pro Plus (Version. 4.0 ) software. The expression of CD127 on thymocytes in aged mice were determined by indirect immunofluorescence flow cytometry.Re sults There were no difference with the body weight, thymus weight and thymus indices among the groups, but the expression of IL 7 mRNA and its receptor in thymocytes were increased in high dose of Salmon silt DNA supplemented group( P <0.05, respectively). Conclusion Our results suggested that supplementation of Salmon milt DNA may postpone age related involution of thymus in Balb/c mice through increasing the production of IL 7 and the expression of its receptor.
