大鼠心肌细胞粘着斑蛋白α-catenin表达的增龄变化

摘要 目的 探讨大鼠心肌细胞发育过程中粘着斑蛋白α -catenin的表达分布方式以及增龄变化规律。方法 应用S -P免疫组化方法分别对大鼠的胎儿、新生儿、幼年、青年、中年和老年大鼠的心肌细胞粘着斑蛋白α-catenin的表达进行检测。结果 细胞粘着斑蛋白α -catenin在大鼠的心房、心室、乳头肌、室间隔等处均有表达。胎鼠心肌细胞中粘着斑蛋白α -catenin分散分布于细胞膜和胞浆中 ;新生大鼠心肌细胞的连接处见有α-catenin的免疫标记颗粒 ;幼年时期α -catenin的免疫标记颗粒大部分集中在心肌细胞末端的闰盘处 ;青年、中年和老年大鼠α -catenin的免疫标记典型的分布在心肌细胞末端的闰盘处。结论 在大鼠心肌发育过程中 ,α -catenin的表达分布存在增龄变化 ;粘着斑蛋白α -catenin的分布方式与闰盘的发育是一致的。 Objective To research the spatiotemporal patterning and age related changes of α catenin expression during development of rat cardiomyocytes. Method Using immunohistochemical method,myocardial α catenin distribution was investigated in fetal rat and postnatal development(1 postnatal day to old rat). Results The expression of α catenin was located in myocardium of atrial and ventricular and septum interventriculare and muscle papillares.The α catenin immunolabeling was distributied in myocyte cell membranes and within cytoplastic in fetal rat heart . In neonatal rat myocardium α-catenin immunolabeling was located in the junctions of the myocardial cells. Lots of α catenin immunolabeling was located in the terminal intercalated disk in infant rat myocardium. From young to old rat heart,the classic α catenin was located in transversely orientated intercalated disk. Conclusion The α catenin expression displayed progressive age related changes and the changing pattern of α catenin is closely related with development of the intercalated disk in rat myocardium.
