摘要
目的 探讨中药雄黄对乳腺癌多药耐药细胞系 (MCF 7/ADM )的逆转作用及可能的逆转机制。方法 药物敏感试验采用四氮唑蓝 (MTT)比色法 ,细胞内药物浓度测定采用荧光分光光度法 ;基因表达水平检测采用RT -PCR法。结果 雄黄在 0~ 2 5 μg/ml浓度范围内对MCF 7/ADM细胞未见明显毒副作用 ;15 μg/ml、2 5 μg/ml雄黄逆转倍数分别为 2 0倍和 2 8倍 ,并能明显抑制mdr 1基因的转录水平。 结论 雄黄可以逆转MCF 7/ADM细胞的多药耐药性 ,并呈剂量依赖性 ;可能的逆转机制为下调mdr
Objective To study the reverse effects and mechanism of realgar on the multidrug resistance of human breast cancer cell line MCF 7/ADM in vitro. Method Cytotoxic effects were determined with MTT colorimetry, intracellular accumulations of adriamyclin (ADM) were determined by immuno spectrophotometry, the expression of ABC gene family was determined by RT PCR.Results Realgar in the concentration range of 0~25μg/ml had no toxic effects on the cells of human treast cancer cell line MCF 7/ADM. When the concentration of realgar increased to 15μg/ml and 25μg/ml, the reverse effects of realgar on the multidrug resistance of human breast cancer cells were increased by 2.2 and 3.0 fold, respectively. Intracellular accumulations of ADM in MCF 7/ADM cells was obviously increased after treatment with realgar, and similar to that of sensitive cells under 25 μg/ml realgar. Realgar can down regulate the expression of mdrl. mrp and bcrp in MCF 7/ADM cells. Conclusion Realgar is able to reverse the multidrug resistance of the cells of human breast cancer cell line MCF 7/ADM in a dosage dependent manner by down regulating the expression of ABC gene family.
