CpG ODN增强免疫力低下小鼠对乙肝疫苗的体液免疫应答 被引量:3

摘要 目的 探讨CpGODN对免疫力低下小鼠的体液免疫增强作用。方法 选用老年C57BL/ 6小鼠和免疫抑制模型小鼠 ,将重组HBsAg疫苗和CpGODN同时或单独肌注到小鼠体内 ,2周后以同样剂量加强免疫 1次 ,再过 3周后摘除眼球取血 ,用ELISA方法检测抗HBsAgIgG抗体。 结果 免疫抑制模型中同时注射CpGODN和疫苗组产生的IgG抗体绝对量是单独注射疫苗组的 2倍 ;老年鼠组中 1 0 μg和2 0 μgCpGODN与疫苗同时注射组产生的IgG抗体绝对量分别是单独注射疫苗组的 3倍和 4倍。 Objective To investigate adjuvant action of CpG ODN i n low immunity mice. Methods Using C57BL/6 aged mice and immunity su ppression model mice, all mice were given an injection into the left tibialis an terior muscle of recombinant HBsAg or both recombinant HBsAg and CpG ODN, then t he mice were given an another injection at 2 weeks using the same formulation. T he mice blood was drawn; eyes were extracted at 5 weeks after immunization and r ecombinant HBsAg specific antibody IgG was measured by ELISA method. Resul ts There was an two-fold increase in anti-HBs IgG when CpG ODN was mixed with recombinant HBsAg(P<0 05) in immunity suppression model mice; there were also three-fold and four-fold increases in anti-HBs IgG when 10 μg and 20 μg CpG ODN were mixed with recombinant HBsAg(P<0 05) in aged mice. Conclusion CpG ODN can enhance the humoral immune responses against hep atitis B vaccine in low immunity mice .
