摘要 目的 研究野生型p5 3基因对人未分化胃癌细胞系HGC2 7生长的抑制作用。方法 用表达人野生型p5 3基因全长cDNA的真核载体pcDNA3 p5 3,转染人未分化胃癌细胞系HGC2 7,对转染后细胞进行生长曲线、细胞生长抑制率及流式细胞术分析 ,观察其生物学特性变化。结果 外源野生型p5 3基因在HGC2 7细胞内稳定表达 ,经pcDNA3 p5 3转染的HGC2 7细胞生长速度受到明显抑制 ,流式细胞分析显示G1期增加 ,S期下降 ,细胞出现凋亡。结论 野生型p5 3基因能在HGC2 7细胞内表达 。 Objective To study inhibitory effects of wild-type p53 gene transfection on the growth of human gastric carcinoma cells HGC27.Methods Human gastric carcinoma cells HGC27 were transfected by an eukaryotic expression vector pcDNA3 containing wild-type p53 genes by lipofectamine.Then,cell growth curve,cell growth inhibition rate and flow cytometry were analysed,and the changes of their biologic characteristics were observed.Results Exogenous wild-type p53 was successfully transfected into HGC27 cells and stable expression was obtained.The growth rate of transfected cells was obviously inhibited.The percentage of phage G 1 cells increased and phage S cells decreased,and the apoptosis was observed by flow cytometry.Conclusion Exogenous wild-type p53 gene could inhibit the proliferation of HGC27 cells and induce the apoptosis of HGC27 cells.
