摘要
目的 :探讨共刺激分子B7 1(CD80 )及MHC分子在急性白血病 (acuteleukemia ,AL)中的表达 .方法 :应用一组mAb ,采用免疫荧光法对AL 5 2例瘤细胞及 34例正常人骨髓单个核细胞 (BMMC)表面B7 1及主要组织相容性复合体(majorhistocompatibilitycomplex ,MHC)分子的表达进行了检测 .结果 :所有患者瘤细胞及正常人BMMC上MHCⅠ类分子表达强阳性 ,MHCⅡ类分子表达 92 %阳性 ,而B7 1表达差异较大 ,急性单核细胞白血病 (M 5 )表达阳性率最高 (8/11) ,而急性粒细胞白血病 (M 1,M2 ,M3)均无表达 .结论 :B7 1缺陷是白血病细胞逃避宿主免疫监视的重要原因 ,转染B7
AIM: To explore the expressions of costimulatory molecule B7 1(CD80) and MHC molecules in acute leukemia (AL). METHODS: With a group of monoclonal antibodies (MoAbs) and by direct or indirect immunofluoressence, the expressions of B7 1 and MHC molecules on the surface of hematologic malignant tumor cells in 52 cases of AL and bone marrow mononuclear cells (BMMC) in 34 healthy persons were detected. RESULTS: All samples were strongly positive (100%) for MHC Ⅰ class molecule. The positive expression rate of MHC Ⅱ class molecule was 92%. B7 1 molecule expression was highest (8/11) in acute monocytic leukemia (M5), but deficient in acute myelogenous leukemia (M1, M2, M3). CONCLUSION: Deficiency of B7 1 is an important cause for leukemic cells to evade host immunosurveiliance and may play an important role in the pathogenesis of AL.
