摘要
目的:研究多奈哌齐对Aβ25-35蛋白诱导的PC12细胞损伤的保护作用。方法:采用细胞培养、MTT法和流式细胞仪技术,以β淀粉样蛋白(amyloidbetaprotein,Aβ)诱导PC12细胞损伤作为细胞模型,观察不同剂量Aβ25-35蛋白对PC12细胞的影响以及多奈哌齐对PC12损伤的保护作用。结果:PC12细胞对于Aβ25-35造成的损伤和凋亡呈剂量依赖型,凋亡百分率增加(71.3%,90.1%),细胞增殖活性降低(t=13.89,P<0.001);提前给予多奈哌齐保护的PC12细胞凋亡百分率明显降低(52.0%),细胞增殖活性较前提高(t=12.77,P<0.001)。结论:多奈哌齐明显减少Aβ诱导的PC12细胞的损伤,提高细胞的存活率,减少细胞凋亡。
AIM:To study the protection of donepezil against PC12 cells injury induced by amyloid beta protein(Aβ) 25-35. METHODS:PC12 cells injury induced by Aβ25-35 were taken as the cell models by using cell culture, MTT assays and flow cytometry. The effect of Aβ25-35 of different dosages on PC12 cells and the protective effect of donepezil on PC12 injuries were observed. RESULTS:The injury and apoptosis of PC12 induced by Aβ25-35 were dependent on dosage, apoptosis percentage increased(71.3%,90.1%), cell proliferation activity reduced(t=13.89, P < 0.001). The apoptosis percentage of PC12 cells, which were protected by donepezil in advance reduced significantly(52.0%), while the cell proliferation activity increased(t=12.77,P < 0.001). CONCLUSION:Donepezil can reduce the injury of PC12 cell induced by Aβ25-35, increase the survival rate and reduce the apoptosis.
