摘要
目的:就电针预处理对大鼠局灶性脑缺血的影响进行实验研究。方法:采用SD大鼠(体质量280~340g,39只)作为研究对象,电针预处理大鼠百会、大椎穴,电针时间分别为15,30,60,90,120min,间隔1h后,造成局灶性脑缺血。行苏木精-伊红和末端脱氧核苷酸转移酶(TdT)介导的脱氧尿苷三磷酸(dUTP)切口末端标记(TUNEL)染色,测定大鼠脑梗死灶面积百分比和脑细胞凋亡情况。结果:电针预处理组中除15min组外,大鼠脑梗死灶面积百分比都比缺血对照组减小,脑细胞凋亡数也减少。预处理60min组脑梗死灶面积百分比和18700.2μm2凋亡细胞数分别为(27.8±4.2)%和(20.2±3.0)个,与缺血对照组犤(41.0±7.6)%和(31.0±4.4)个犦比较差异有显著性意义(P<0.05)。结论:电针预处理能够保护随后的缺血性脑损伤,且预处理需要达到一定的时间才具有保护作用。
AIM:To study the effect of electroacupunture(EA) preconditioning on focal cerebral ischemia in rats. METHODS:Thirty nine Sprague Dawley rats weighing 280-340 g were selected as experimental objects.Paihui(GV 20) and Tachui(GV 14) of the rats were stimulated with EA for 15,30,60,90 and 120 minutes respectively.After one hour,those rats' cerebral middle artery was occluded by a 4-0 monofilament nylon suture to cause focal cerebral ischemia.Using Hemotoxin and Eosin(HE) and terminal deoxynucleotidy transferase mediated d UTP nick end labeling (TUNEL) staining methods,the infarct area percentage and cerebral apoptosis were measured. RESULTS:All the cerebral infarct area percentage and the cerebral apoptosis of EAP groups except EAP group of 15 minutes were reduced or decreased as compared with the control group. The infarct area percentage and cerebral apoptosis of EAP group of 60 minutes were(27.53±5.39)%and(20.2±3.0) per unit area(P< 0.05),significantly higher than those of the control group[(41.0±7.6)%and (31.0±4.4) per unit area](P< 0.05),respectively. CONCLUSION:Electroacupuncture preconditioning can protect brain injury following cerebral ischemia,though enough time of EAP is needed.
