还元注射液对实验性脑出血大鼠脑组织显微、超微结构的影响(英文) 被引量:6

摘要 目的:探讨还元注射液对实验性脑出血大鼠脑组织显微、超微结构的影响及保护作用。方法:将30只SD大鼠随机分为正常组、模型组和还元组,每组10只。各组于造模后2h分别腹腔注射生理盐水和还元注射液1mL/(100g.次),2次/d,共给药3d。第4天断头处死迅速取脑,制成光镜、电镜片,观察组织学变化。结果:①正常组:光镜、电镜下脑组织结构正常;②模型组:光镜下,血肿范围大,充满变性红细胞,脑组织变性、坏死,毛细血管严重水肿;电镜下,神经细胞和胶质细胞明显肿胀,胞浆内大部分细胞器消失,染色质消失,神经细胞脱髓鞘,轴浆消失;③还元组:光镜下,血肿形成囊肿,与周围脑组织界限不十分明显,血肿内有大量的胶质细胞和巨噬细胞,血肿周围无明显变性神经细胞,毛细血管无明显水肿;电镜下,神经细胞和胶质细胞无明显肿胀,胞浆内细胞器结构基本完整,神经纤维结构大致正常。结论:还元注射液对实验性脑出血大鼠脑组织结构具有保护作用。 AIM:To explore the effect of Huanyuan injection on the microstructure and ultrastructure of the cerebral tissues in rats with experimental cerebral hemorrhage. METHODS:Thirty SD rats were randomized equally into normal control group,cerebral hemorrhage model group and Huanyuan injection treatment group,to receive treatment with intraperitoneally injection of saline(in the former 2 groups) or Huanyuan injection(10 mL/kg,twice a day for 3 d) as indicated 2 h after cerebral hemorrhage.On the fourth day,the rats were sacrificed and the brain sections were prepared for observing the histological changes through optical and electron microscopy. RESULTS:In the model group,the rats had extensive hematoma filled with degenerated erythrocytes,and brain tissue degeneration,necrosis,and severe edema of the capillaries could be observed microscopically.Under electron microscope,the neurons and spongiocytes were obviously swollen,the majority of the organelles in the cytoplasm and chromatin in the nuclei disappeared,with demyelination of the nerve fibers and absence of the axoplasm.In the Huanyuan group,a cyst was formed around the hematoma with no obvious boundary with the surrounding tissues.A large number of spongiocytes and macrophages were present in the hematoma,and no obvious degeneration of the neurons or edema of the capillaries was observed.Under electron microscope,the neurons and spongiocytes presented no obvious degeneration,and the structures of the organelles and nerve fibers were basically normal. CONCLUSION:Huanyuan injection can protect the structures of the neurons in the brain tissue in rats with experimental cerebral hemorrhage.
