摘要
目的:探讨镁离子(Mg2+)在0.1mmol/L谷氨酸酸诱发的海马神经元损伤中的作用。方法:将神经元从新生SD大鼠海马中分离后培养6~9d,即单纯随机分成3组。对照组:单纯培养基;谷氨酸组:培养基+5种不同浓度的谷氨酸;镁离子组:培养基+5种不同浓度的Mg2+,30min后再加入0.1mmol/L谷氨酸。结果:①谷氨酸组海马神经元存活率与对照组相比显著地呈剂量依赖性降低,谷氨酸的剂量为2mmol/L时细胞存活率为4.64%。②与谷氨酸组比较,镁离子组低浓度的Mg2+可提高海马神经元的存活率,4.5mmol/LMgSO4处理海马神经元的存活率比谷氨酸组增加78.41%。2+结论:低浓度的Mg能保护谷氨酸诱发损伤的海马神经元。
AIM:To investigate the effect of magnesium ion(Mg2+) on neural damage in the hippocampus induced by 0.1 mmol/L glutamic acid. METHODS:The neurons isolated from the hippocampus in rats were cultured for 6-9 d,and then randomly divided into three groups:A.medium alone;B.medium+glutamic acid(5 different levels);C.medium+Mg2+(5 different levels),and then 0.1 mmol/L glutamic acid was added into them 30 min later. RESULTS:①Compared with that of group A,the survival rate of hippocampal neurons in group B was remarkably decreased in a dose dependent manner.②In contrast with group B,the survival rate of hippocampal neurons in group C was significantly increased when the concentration of Mg2+was lower. CONCLUSION:Mg2+in a lower concentration can protect hippocampal neurons against damage induced by glutamic acid.
