摘要
目的 检测缺氧诱导因子 - 1(HIF- 1)和血管内皮生长因子 (VEGF)与结直肠癌临床病理及预后的关系。方法 用免疫组化 SP法检测 6 1例结直肠癌组织中 HIF- 1α及 VEGF蛋白的表达。结果 结直肠癌组织中HIF- 1α蛋白呈高表达 ,阳性率为 6 5 .6 % ,癌旁及正常组织不表达 ,HIF- 1α与结直肠癌的浸润深度及淋巴结转移密切相关 (P<0 .0 1) ,与 Dukes分期呈正相关 (P<0 .0 1) ;结直肠癌 VEGF阳性表达率为 4 9.2 % ,癌旁及正常黏膜几乎不表达 VEGF,癌间质血管内皮细胞不表达或弱表达 ,VEGF与结直肠癌的浸润深度及淋巴结转移相关(P<0 .0 5 ) ,与 Dukes分期呈正相关 (P<0 .0 2 ) ,且 VEGF阳性表达与 HIF- 1α阳性表达程度呈正相关 (P<0 .0 0 1)。结论 HIF- 1α与 VEGF在结直肠癌浸润、转移过程中有协同作用 ,同时检测 HIF- 1α、VEGF蛋白的表达可作为判断结直肠癌浸润。
Objective To explore the relationship between the hypoxia-inducible factor-1α(HIF-1α)and vascular endothelial growth factor(VEGF) and the clinical pathology and prognosis of colorectal carcinoma.Methods The expression of HIF-1α and VEGF in 61patients with colorectal carcinoma were tested by the SP methods of immunohistochemistry.Results The expression of HIF-1α protein was higher in patients with colorectal carcinoma,the positive rate was 65.6%,and was no expression in tissue around the tumor and normal tissue,close relationship was found between HIF-1α and invasion depth and lymph node metastasis,P<0.01.HIF-1α had postive correlation with Dukes stages,P<0.01.The positive rate of VEGF was 49.2%;there nearly was no VEGF expression in tissue around the carcinoma and normal tissue,was no or minor expression in vascular endothelial cells of carcinoma Interstitial tissue;close correlation was found between VEGF and the invasion depth?lymph node metastasis of colorectal carcinoma,P<0.05.VEGF had positive correlation with Dukes stages,P<0.02,there was postive correation between VEGF postive expression and HIF-1α positive expression dogrees,P<0.001.Conclusion There is synergistic action between HIF-1α and VEGF in the invasion and metastasis in colorectal carcinoma.HIF-1α,VEGF which are tested at the same time can be tested as an important indicator to judge those biological behavior of colorectal carcinoma such as invasion,metastasis and prognosis.
