MMP-2、MMP-7与胃癌的侵袭转移关系探讨

Relationship between matrix metalloproteinase and metastasis of gastrtic cancer

目的 观察基质金属蛋白酶 (MMP)系列中的 型胶原酶 (MMP- 2 )和基质溶解素 (MMP- 7)在胃癌和癌旁组织中的表达 ,探讨其与胃癌侵袭转移的关系。方法 采用逆转录 - PCR(RT- PCR)方法分别检测 4 0例胃癌及癌旁组织 MMP- 2和 MMP- 7m RNA的阳性表达及含量。结果 4 0例胃癌组织中 MMP- 2和 MMP- 7的阳性表达率分别为 82 .5 %、72 .5 % ,明显高于癌旁组织的 72 .5 %、17.5 % ,二者比较差异有显著性 (P<0 .0 0 1)。MMP-2和 MMP- 7表达量高于其癌旁组织 (t=3.6 7,t=7.894 ) ,差异有显著性 (P均≤ 0 .0 0 1)。MMP- 2和 MMP- 7的表达量与胃癌患者的性别、年龄、肿瘤的大小均无相关性 (P>0 .0 5 )。 MMP- 2的表达量在晚期、低分化及伴有淋巴结转移的胃癌患者中明显高于早期、高分化及无淋巴结转移的胃癌患者 (P<0 .0 5、P<0 .0 1)。 MMP- 7在各期胃癌中的表达无差异 (P>0 .0 5 ) ,但在中低分化及合并有淋巴结转移的胃癌患者中其表达量明显高于高分化及不伴有淋巴结转移的胃癌患者 (P<0 .0 5 )。结论 MMP- 2和 MMP-

Objective Objectlve To study the expression Of matrix metalloproteinases(MMP)in gastric cancer and their corresponding adjacent tissues,to investigate the possible mechanism of tumor lnvasion and metastasis in gastric cancer.Method Tumor tissues and adjacent tissues in 40 cases of gastric cancer were detected by the reverse transcriptase-polymerase chain reaction(RT-PCR).Results The expression of MMP-2 and MMP-7mRNA in tumor tissues were higher than that in adjacent tissues,respectively(P≤0.001).The expression was related to tumor size,degree of differentiation and clinicai staging.The expression of MMP-2 and MMP-7mRNA inl7gastric cancer with lymph node metastasis were higher than that in without lymph node metastasis.Conclusion MMP-2 and MMP-7 are related to tumor invasion and metastasis in gastric cancer.
