摘要
目的观察丝裂霉素 (mitomycin ,MMC)、5 氟尿嘧啶 (5 fluorouracil,5 Fu)、三苯氧胺(tamoxifen ,TAM)联合应用对人乳腺癌细胞株MCF 7和MDA MB 4 35S的作用。方法采用MTT法 (四甲基偶氮唑蓝比色法 ,3 (4,5 dimethyl 2 thiazoly) 2 ,5 diphenyl 2H tetrazolium bromide)利用中效原理判断药物联合应用的效应。结果MMC、5 Fu单用及联合应用时 ,随着药物浓度增加其效应也增加 ;TAM与MMC、5 Fu合用时 ,TAM +MMC中效浓度为 1 6 5× 10 -5mmol/L ,TAM +5 Fu中效浓度为 0 0 32 6mmol/L ,对MCF 7细胞作用加强 ,而对MDA MB 4 35S细胞作用不明显 ;MMC与5 Fu高浓度合用时呈协同作用 (CI <1) ,低浓度时呈拮抗作用 (CI >1) ;两药合用时浓度比例变化及给药时间次序对效应影响不大。结论TAM与MMC、5 Fu合用时 ,对MCF 7细胞作用加强 ,而对MDA MB 4 35S细胞作用不明显。
ObjectiveTo evaluate the killing effects of MMC,5-Fu and TAM on human breast cancer cell lines (MCF-7 and MDA-MB-435S). MethodsThe MTT method and median-effect principle were used. ResultsKilling effects of drugs were enhanced as drug concentration increased while they were used alone or in combination. The median-effect concentration of TAM in conjunction with MMC was 1.65×10 -5 mmol/L,and that of TAM in conjunction with 5-Fu was 0.032 6 mmol/L. MMC was synergetic with 5-Fu at high concentration(CI<1) and of antagonist with 5-Fu at low concentration(CI>1). The ratios of drug concentration and the sequence of administration were not related with the killing effects. ConclusionsA combination of MMC, 5-Fu and TAM has an increased killing effect on MCF-7 cell lines and questionable enhanced effect on MDA-MB-435S cell lines.
