摘要
目的 探讨永久性脑缺血神经干细胞 (NSC)迁移是否伴随自身的分裂与增殖。方法 以永久性脑缺血大鼠为模型 ,采用免疫组化染色方法 ,观察脑缺血 1,3,7,14和 2 8dNSC迁移及增殖细胞核抗原(PCNA)的表达。结果 室管膜下区 (SEZ)NSC和PCNA+ 细胞在缺血后沿胼胝体腹侧向缺血区方向发生了时空一致性的迁移模式 ;缺血梗死灶周围均有较多巢蛋白和PCNA阳性细胞的表达。结论 永久性脑缺血后NSC可能通过自身分裂和增殖向外迁移 ,巢蛋白和PCNA在缺血区周围表达可作为NSC迁移和增殖的标志物。
Objective To understand whether the migration of neural stem cells(NSCs) accompanies themself’s cleavage and proliferation after permanent cerebral ischemia. Methods Using rat’s permanent cerebral ischemia model, immunohistochemical staining was used to observe the migration of NSCs and the changes of the expression of nestin and proliferation cell nuclear antigen(PCNA). Results The NSCs and PCNA cells presented a migration pattern with time space accordance from the subependyma zone(SEZ) to the ischemic zone along the ventral side of the corpus callosum after ischemia, much nestin cells and PCNA cells were found surrounding the ischemic area successively after ischemia. Conclusion The NSCs in SEZ are likely to migrate from SEZ to the ischemic zone accomanying cell cleavage and proliferation for which the expression of nestin and PCNA near the ischemic zone may be the marker.
