摘要
目的:研究培养的人骨髓间充质干细胞(MSCs)与人发角蛋白(HHK)人工腱的相容性以及MSCs在HHK上的生长情况。方法:人MSCs与HHK人工腱联合培养后,分别在倒置显微镜及扫描电镜和透射电镜下观察MSCs的生长状态和超微结构。结果:MSCs在光镜下呈单层或多层贴附于HHK人工腱表面,扫描电镜下呈细长梭形,有的呈扁平多角形,其生长方向与HHK人工腱长轴相一致。透射电镜下可见静息态和活动态两种不同功能状态。结论:MSCs与HHK人工腱之间有极好的粘附性和相容性,是一种理想的组织工程种子细胞。
Objective : To investigate the compatibility between human marrow-derived mesenchymal stem cells (MSCs) and human hair keratin (HHK) artificial tendon in vitro. Methods : The co-cultured human MSCs and HHK artificial tendon were studied using inverted microscope and scanning electron microscope; the ultrastructure of MSCs was observed under transmission electron microscope. Results : Under inverted microscope, the single or multi-layer MSCs adhered to the surface of each hair of HHK artificial tendon. Most MSCs growing on the surface of HHK artificial tendon had long spindle; some were flat and multi-angle in shape. Static MSCs and active MSCs could be identified under transmission microscope. Conclusion : MSCs have good compatibility with HHK artificial tendon, and the MSCs are ideal seed cells for tissue engineering.
