大鼠脑出血后血肿周围组织线粒体ATPase-6基因表达及TUNEL阳性细胞动态变化研究 被引量:2

摘要 目的 研究脑出血后脑组织线粒体 ATPase- 6基因表达变化与细胞凋亡的相关性。方法 以 5 0μl鼠尾血注入大鼠尾状核制作大鼠脑出血模型 ,注血后分别于 12 h、1d、3d、7d处死大鼠 ,断头取血肿周围脑组织。 (1)用 RT- PCR方法测定以上不同时相点大鼠脑出血后血肿周围水肿组织线粒体 ATPase- 6基因表达情况 ;(2 ) Td T介导的 d UTP缺口末端标记法检测 TU NEL阳性细胞的表达。结果 出血后 12 h、1d、3d线粒体 ATPase- 6基因表达较对照组下降 ,但差异不显著 (P>0 .0 5 )。至第 7天时表达明显下降 (P<0 .0 1)。 TU NEL阳性细胞在出血后12 h开始表达 ,3d达到高峰 ,7d时仍持续较高水平。结论 脑出血早期血肿周围水肿组织线粒体 ATPase- 6基因表达无明显减少 ,晚期表达明显下降 。 Objective To investigate the expression changes of ATPase 6 gene of cerebral mitochondria and TUNEL positive cells after intracerebral hemorrhage(ICH) in rats.Methods Rat ICH model was established with 50μl tail blood injected into caudoputamen ,while nothing was injected in control.Rats were killed respectively at 12h,1d,3d and 1w after injection.The expression of ATPase 6 gene in rat cerebral mitochondria around the hematom was measured by RT PCR,while the expression of TUNEL positive cells by terminal deoxynucleotidyl transferase mediated dUTP nick end labelling.Results At 12h,1d,3d after injection,the expression of ATPase 6 gene didn't decrease obviously than that in the control group ( P >0.05).But at 7d after injection,it decreased markedly( P <0.01).TUNEL positive cells were detected at 12h after ICH,peaked at 3d,and lasted at 7d.Conclusion The expression of ATPase 6 gene changed slightly in the early time after hemorrhage,but it decreased markedly later,while TUNEL positive cells were detected correspondingly in the early time.
