摘要
目的 :寻找一种安全、可靠、皮内存留时间最长 ,疗效最佳的注射材料 ,以达到更好地抗皮肤老化的目的。方法 :健康SD大白鼠 2 4只 ,随机分成 4组。在大白鼠背部用纹眉液作 6个直径 1cm的圆圈标记 ,为A~F 6组。先取大白鼠颈部皮肤制成真皮微粒 ,然后将自体真皮微粒 (A组 )、自体真皮微粒加透明质酸钠混悬液 (B组 )、生理盐水 (C组 )、透明质酸钠 (D组 )、胶原(E组 )、自体真皮微粒加透明质酸钠和胶原混悬液 (F组 )均制成 0 .3ml混悬液 ,分别注射至大白鼠背部标记区真皮内。术后 1周、4周、12周、2 4周 ,随机抽取 1组 ,切取大白鼠背部皮肤组织 ,作常规病理检查观察皮肤的改变 ,胶原增生的情况和皮肤的厚度及超微结构检查 ,测量结果采用SAS软件作统计学处理。结果 :巨体标本观察 :注射术后 2 4周仅F组局部可见有注射物存留。采用自体真皮微粒、透明质酸钠、胶原皮内注射均可使大白鼠背部局部皮肤增厚。采用自体真皮微粒加透明质酸钠和胶原 3种材料混悬液有增强效应 ,局部皮肤厚度增加最明显 ,与各对照组比较有显著差别 (P <0 .0 1)。结论 :采用自体真皮微粒加透明质酸钠和胶原 3种材料混悬液局部皮内注射 ,可延长注射材料皮内留存的时间 ,使吸收率降低 ,局部皮肤明显增厚 ,达到更好地抗皮肤老化的作用。具?
Objective: To search a safe, reliable and effective injectable materials for the aging skin augmentation. Methods: Twenty four 6-month-old SD rats, were used in this study. A piece of 3 cm ×1 cm full thickness skin on the neck of each rat was harvested to be minced into 0.3~1 mm particles by scissors. There were 6 different kinds of injectable materials used in this study: 1) autologous dermal particles (ADP) (Group A), 2) the mixture of ADP and natrium hyaluronate (Group B), 3) saline (Group C), 4) natrium hyaluronate (Group D), 5) collagen (Group E), and 6) the mixture of ADP, natrium hyaluronate and collagen (Group F). Six 1 cm diameter circles were tattooed on the dorsal skin of each rat. Each of 6 kinds of injectable materials were injected into the dermis in turn of the marked skin circles on the rat. Six rats were sacrificed randomly at 1, 4, 12, and 24 weeks after injection. The specimens were harvested after macroscopic assessments, and then subjected to histological analysis. Part of these specimens were also sent to microstructure assessments by electron microscope. The measurements of the thickness of the dermis underwent statistic analysis of SAS system. Results: Macroscopic assessments showed only part of the materials of Group F could be found at 24 weeks after injection. The dorsal skin was thicker by injecting ADP, natrium hyaluronate or co1llagen in comparison with injecting saline (Group C). The dorsal skin was thickest by injecting the mixture of ADP?natrium hyaluronate and collagen(Group F) among these six groups at each of different times. There was statistically significant difference between each two groups (P<0.01). Conclusion: It suggested that endodermal injection of the mixture of ADP, natrium hyaluronate and collagen (Group F) could prolong the remaining time of the injectable materials. The results of the skin augmentation were the best in this experimental group. It revealed the value for the clinical applications.
