丙丁酚加维生素E对高脂饮食大鼠肾间质纤维化的影响 被引量:3

摘要 目的 :观察抗氧化治疗 (丙丁酚 +VitE)对高脂饮食大鼠肾间质纤维化程度及其所引起的肾细胞因子表达的影响。方法 :单肾切除大鼠分为对照组 (1 0只 ,正常大鼠饲料 )、高脂饮食组 (1 0只 ,大鼠饲料含质量分数 1 .5 %胆固醇、0 .5 %胆酸、5 %动物油 )、治疗组 (1 0只 ,高脂饮食加丙丁酚 5 0 0mg/d、VitE 5 0IU/d)。治疗 1 2周后测定肾功能、肾组织MDA含量 ,观察肾脏病理切片中间质病变程度 ,免疫组化方法检测肾组织胶原I和TGF - β1 的表达。结果 :与高脂饮食组相比 ,治疗组肾功能改善 ,肾组织MDA含量降低 ,肾间质病变减轻 ,肾组织胶原I和TGF - β1 的表达减弱。结论 :抗氧化剂可减轻高脂饮食大鼠肾间质纤维化的程度 ,同时抑制肾间质TGF - β1 的表达。 Aim: To investigate whether antioxidant therapy (probucol+VitE) could attenuate interstitial fibrosis in high-lipid diet rats and to observe the changes of renal cytokine expression. Methods: A left nephrectomy on thirty female Sprague-Dawley rats was performed. The rats were randomly allocated into three groups, normal control(ten animals, normal rat's forage), high-lipid diet group (ten animals, 1.5% cholesterol+0.5% cholic acid+5% animal oil), treatment group (ten animals, high-lipld diet, probucol 500 mg/d+VitE 50 IU/d). After 12 weeks renal function and MDA were measured. The expression of collagen I and transforming growth factor-β 1 (TGF-β 1) were examined by immunohistochemistry. Results: Compared with the high-lipid diet group, the renal function ameliorated and the level of MDA in renal interstitium was significantly lower in the treatment group( P <0.01). The expressions of collagen I and TGF-β 1 in the treatment group were lower than those in the high-lipid diet group( P <0.01). Conclusion: Probucol plus VitE can inhibit the expression of TGF-β 1 in high-lipid diet rats. It is suggested that antioxidant therapy reduces the degree of renal interstitial fibrosis in high-lipid diet rats by down-regulating TGF-β 1 expression.
