碧萝芷对老年大鼠的抗氧化作用

摘要 目的 :探讨碧萝芷对老年大鼠的抗氧化作用。方法 :选用 4 0只 2 0月龄的Wistar大鼠 ,分为空白对照组及碧萝芷高、中、低剂量组 4组 ,分别给予蒸馏水、受试物 5 0 .0mg/kg、1 6 .7mg/kg、8.4mg/kg每d灌胃 1次 ,连续 8周。采用黄嘌呤氧化酶法测血清、肝及脑组织匀浆的SOD活性 ,硫代巴比妥酸 (TBA)法测血清及肝、脑组织中MDA含量 ,并同时检测脑组织的单胺氧化酶 (MAO)活力和脂褐质含量。结果 :与空白对照组比较 ,碧萝芷各剂量组血清、肝、脑组织中的MDA含量均较低 (P <0 .0 5 ) ,除肝组织外SOD的活性明显升高 (P <0 .0 5 ) ;脑组织MAO活力及脂褐质含量也均低于空白对照组。各剂量组间比较 ,高剂量组脑组织MDA含量高于低剂量组 (P <0 .0 5 ) ,中、高剂量组脑组织MAO活力及脂褐质含量低于低剂量组 (P <0 .0 5或P <0 .0 1 )。结论 :碧萝芷有抗氧化作用 ,可延缓大鼠衰老进程。 Aim: To study the antioxidant effects of pycnogenol on aged rats. Methods: Forty male Wistar rats were randomly allocated into four groups (normal control, low dose, mediate dose, and high dose groups) and given 0.0, 50.0 mg/kg, 16.7 mg/kg, 8.4 mg/kg pycnogenol once per day for 8 weeks. The activities of SOD in serum, homogenate of liver and brain were measured by the method of xanthine oxidase, the contents of MDA in serum, homogenate of liver and brain were measured by the method of thiobarbituric acid(TBA), and activity of MAO and content of lipofuscin in brain were measured in the aged rats. Results: Pycnogenol significantly decreased the content of MDA,and significantly increased the activity of SOD except in liver tissues in experimental groups ( P <0.05). Brain MDA content in high dose group was higher than that in low dose group ( P <0.05),and MAO activities in brain and contents of lipofuscin in mediate dose and high dose groups were lower than those in low dose group( P <0.05 or P <0.01). Conclusion: Pycnogenol can enhance antioxidant activities in aged rats.
