摘要
紫杉醇是一种天然抗癌药物.采用自乳化溶剂蒸发法制备了负载紫杉醇的聚乙二醇/聚己内酯三嵌段共聚物纳米粒(PMT),运用动态光散射(DLS)测定PMT的粒径和粒径分布,并用透射电子显微镜(TEM)表征形态结构.同时,还研究了投药量对PMT的影响及PMT的体外药物释放行为.实验结果表明:PMT的粒径为纳米级,且随着共聚物相对分子质量和载药量的增大而增大;PMT的体外释放无突释现象,释放速率缓慢.
Paclitaxel is a kind of natural anticancer drug. Paclitaxel-loaded poly(ethylene glycol)-b-polycaprolactone-b-poly(ethylene glycol) triblock copolymer nanoparticles (PMT) were prepared by self-emulsification/solvent evaporation method. The average size and size distribution of PMT were determined by dynamic light scattering (DLS) and the structure of PMT was characterized by transmission electron microscopy (TEM). At the same time, the influences of the drug-fed amount on PMT and release behavior in vitro of PMT were investigated.The results show that the size of PMT,which is in nanoscale,increases as the copolymer molecular weight and the drug-loaded amount increase.What′s more,the initial burst release in vitro hasn′t been observed and the drug release rate is low.
