甘薯组织培养高效植株再生体系的建立 被引量:6

ESTABLISHMENT OF HIGH EFFICIENT PLANT REGENERATION SYSTEM ON SWEET POTATO TISSUE CULTURE

摘要 以甘薯的叶片和茎段为外植体,建立了甘薯组织培养高效植抹再生体系。愈伤组织诱导的最适培养基为MS+0.1 mg/L NAA+1.0 mg/LBA,最适的芽诱导培养基为MS+1.0mg/L NAA。在该培养基上芽直接由外植体上产生,有的单生,有的簇生,芽最高诱导率为88.9%。不同基因型问以及同一基因型不同外植体间芽诱导率不同。诱导出的不定根转入新的培养基中,仍能分化出芽。诱导出的芽转入不合任何激素的MS培养基中,20天后长成7~10cm高的小苗,移入无菌土盆中,成苗率达100%。 High efficient plant regeneration system on sweet potato tissue culture was established from leaf and stem culture of sweet potato cv Ning-331、Jinanhong and Qianxi-682 respectively. MS was the basal medium in all treatments, supplemented with diffexent kinds and different concentrations of phytohormones. The best callus induction and growth medium is MS+0.1 mg/L NAA+1.0 mg/L BA which gives callus induction frequency of 100%. The optimal shoot induction medium is MS+1.0 mg/L NAA, on this medium, shoot directly regeneration from leaf and stem explants, but it was variance between different tissue or different genotype, the highest shoot regeneration frequency is 88.9%. The adventitious root was transfered into fresh MS medium without any hormones, it could been regenerated wuole plant rapidly. The regenerants were transplanted into sterilized soil and developed into normal plants.
