以酚酞作指示剂 ,在琼脂固态培养基上 ,根据酚酞变为无色所形成变色圈大小筛选环糊精糖基转移酶高产突变株是一种平板快速筛选方法。嗜碱芽孢杆菌 1177菌株经紫外线和γ射线诱变处理后 .经该法筛选得到突变株圈 7- 12 ,该菌株产环糊精糖基转移酶达 5 6 0 0u/ml,较出发菌株提高 183%。连续 5代传代试验表明 ,突变株 7-
It is a rapid method for screening the cyclodextrin glycosyltransferase producer according to the size of the colorless ring by using phenolphthalein as an indicator in the agar solid medium containing soluble starch.The mutant 7-12 which produces as much as 5600u/ml of cyclodextrin glycosyltransferase was obtained by using the rapid screen method after Bacillus alkalophilus 1177 was mutagenized by UV and γ irradiation with 60Co.The continuous 5 generations test shows that the mutant 7-12 is stable in the cyclodextrin glycosyltransferase production.
