摘要
目的 :通过细胞学实验方法观察加温联合化疗药物 -羟基喜树碱 (HCPT)对人肺腺癌Anip973细胞的影响。方法 :采用MTT法 ,得出不同温度 (37 0℃ ,4 1 0℃ ,4 2 0℃ ,4 3 0℃ )下的药物生物活性曲线 ,并对各组数据用SPSS统计软件进行相关性分析。结果 :同一温度下 ,HCPT浓度越高 ,则细胞存活率越低 ,二者呈显著负相关 ,P <0 0 5 ;同一HCPT浓度下 ,当药物浓度 <6 0 μg mL时 ,随着温度的增高 ,细胞存活率降低 ,二者呈显著负相关 ,P <0 0 5 ;药物生物活性曲线特征参数IC50 值与温度亦呈显著负相关 ,P <0 0 5。结论 :同一温度下 ,羟基喜树碱对人肺腺癌细胞系Anip973的杀伤作用呈显著的浓度依赖性 ,而在一定浓度羟基喜树碱作用下 ,加温治疗与化疗联合作用对人肺腺癌细胞的杀伤作用呈显著的温度依赖性 ,并且证明羟基喜树碱为无阈值药物 。
Objective To increase the therapy rate of cancer,hyperthermia and 10-hydroxycamptothecin (HCPT) were applied in this experiment to investigate the effect of some combination treatments on human lung adenocarcinoma cell line Anip973.Methods MTT and SPSS were used to observe and analyze the effect.Results Along with hyperthermic temperature or HCPT's concentration increasing,cell survival fraction decreased,they were negative correlative,P<0.05,and along with hyperthermic temperature rising,the value of IC 50 dropped,they were also negative correlative,P<0.05.Conclusion hyperthermia and HCPT all can apparently resist and kill Anip973,and hyperthermia can strengthen the function of HCPT that had not threshold,which can be applied in clinical work.
