草莓花药离体条件下形态发生的研究 被引量:6

摘要 以草莓花药为外植体,研究了基本培养基、激素、低温诸因子对小孢子启动,愈伤组织形成及植株再生的影响,并探讨了再生植株的起源和途径。(1)外源激素对小孢子发育的启动并非必要的条件,但对形成胚性多细胞体,进而形成胚性愈伤组织却是必不可少的。在附加IAA(0.2mg/L)和BA(2.0mg/L)的F培养基上,有三种不同的生长反应:形成胚性愈伤组织、再生出植株和一枚花蕾。(2)低温处理能有效地提高愈伤组织的诱导频率和再生植株的能力,而且扩大了形态发生的激素浓度范围。(3)基本培养基影响着小孢子的发育结果。只在附加NAA(0.2mg/L)和BA(2.0mg/L)的F基本培养基上直接形成植株,或形成愈伤组织,再从其产生出植株。(4)再生植株是由小孢子或药壁细胞经胚状体发生途径形成的,或小孢子和药壁细胞形成胚性多细胞体,进而发育为愈伤组织,再经胚状体发生而成。 The effect of hormone,different base media and lower temperature on pollen inimination, calli formation and plant regeneration were studied in strawberry anthers In Vitro. At same time, the origin way of regenerated plant were studied too. 1. The exogenous hormones were not essential for the inimination of pollens development, but absolutely necessary for induction and formation of calli, and embryonic multi- cellular structure.There were three different growth respones on medium F with IAA(0.2mg/L) and BA(2.0mg/L), they were calli formated, plants regenerated and flower bud regenerated. 2 The induction frequency of calli and capacity of plants regeneration were increased,and the level of hormone of morphogenesis were also enlarged by lower temperature (4℃). 3. The way of pollens development was affected by base media, plants were only formated by the way of embryogenesis directly from anthers, or calli formated on medium F with NAA (0.2 mg/L)and BA(2.0mg/L).4.Plants regenerated by the way of embryogenesis originated from pollens and cells of anthers wall, and so did calli.
