摘要
目的 探讨甲状腺肿瘤组织以及同组织细针穿刺标本中端粒酶活性的表达及其临床价值。方法 应用端粒重复序列扩增法结合酶联免疫吸附实验 (TRAP -PCR -ELISA)半定量方法对经病理确诊的 12例甲状腺癌、12例甲状腺腺瘤、8例甲状腺增生性结节、12例正常甲状腺组织标本及其细针穿刺标本中端粒酶活性进行检测。结果 甲状腺癌组织及其细针穿刺标本中端粒酶活性水平和阳性率均显著高于腺瘤、增生性结节和正常甲状腺组织及细针穿刺标本的端粒酶活性水平和阳性率 (P <0 .0 1)。结论 端粒酶活性在甲状腺癌中高度表达提示其可能是一个特异性较强的甲状腺恶性肿瘤标记物 ,甲状腺细针穿刺标本中端粒酶活性的检测有助于手术前鉴别诊断甲状腺瘤的良恶性 。
Purpose: To investigate the expression of telomerase activity in tissues and corresponding fineneedle aspiration(FNA) samples of thyroid neoplasms, and its clinical valuation. Methods: The technique of semi-quantitive and PCR-based telomeric repeat amplification protocol enzyme-linked immunoabsordent assay (TRAP-PCR-ELISA) was employed to detect telomerase activity in tissues and corresponding FNA samples from 12 thyroid carcinomas, 12 thyroid adenomas, 8 thyroid hyperplastic nodules and 12 normal thyroids which were histologically confirmed. Results: The levels and the positive rates of the telomerase activity in thyroid carcinoma tissues and FNA samples were significantly higher than those in tissues and FNA samples of thyroid adenomas, thyroid hyperplastic nodules and normal thyroids (P<0.01). Conclusions: Increased expression of telomerase activity in malignant thyroid neoplasms suggests that it is likely a good malignant thyroid tumor marker. The detection of telomerase activity in the FNA samples of thyroid contributes to the differential diagnosis of benign from malignant thyroid neoplasms before operation and has an instructional significance for the management of thyroid neoplasms.
