摘要
目的 制备抗HER2 /neu的特异性单克隆抗体 ,筛选出能够分泌抑制 p185HER2 /neu蛋白过表达的肿瘤细胞生长的单抗杂交瘤细胞株。方法 原核表达含全长neu蛋白的融合蛋白 ,免疫Balb/c雌性小鼠后取效价高的小鼠尾静脉加强后行脾细胞与SP2 / 0细胞进行细胞融合。以ELISA、免疫细胞化学、免疫组织化学、细胞免疫荧光和MTT法进行筛选。结果 共获得 8株稳定分泌HER2 /neu单抗的杂交瘤细胞株 ,其中 5株细胞产生的抗体体外实验能够抑制乳腺癌细胞HBT133的生长。结论 能够抑制HBT133细胞生长的 5株杂交瘤细胞株可以用作人源化 。
Purpose: To prepare specific anti-HER2/neu monoclonal antibody, and select clones which can inhibit the growth of HER2/neu overexpressing tumor cells. Methods: The HER2/neu protein expressed by Prokaryotic vector was used to immunize female Balb/c mice. After cell fusion, ELISA, immunocytochemistry, immunocytofluorescence, immunochemistry and MTT were used to select clones. Results: Five clones that could inhibit the growth of HBT133 breast tumor cells were selected from 8 hybridoma cell lines which can consistently produce anti-HER2/neu monoclonal antibodies. Conclusions: These 5 hybridoma cell lines can be used for humanization which may be very useful in the therapy of breast cancer.
