3-异丁基-1-甲基黄嘌呤和CPT-环磷腺苷促进视网膜节细胞再生的机制探讨 被引量:1

摘要 目的 用不同的抑制剂通过抑制不同的传导途径 ,探讨 3 异丁基 1 甲基黄嘌呤 (IBMX)和CPT 环磷腺苷 (CPT cAMP)促进RGCs再生的可能机制。 方法 采用逆行示踪标记术和定位解剖学技术观察对照组和各实验组的视网膜节细胞再生情况。 结果  1 对照组 1(AG)术后 4周 ,视网膜再生的节细胞平均数为 14 2 8± 2 84个 视网膜 ;2 对照组 2 (AG +IBMX +cAMP)术后 4周 ,视网膜再生的节细胞平均数为 4 917± 14 89个 视网模 ;3 各实验组在上述时间点视网膜再生的节细胞平均数分别为 :(1)H89组 (AG +IBMX +cAMP +H89) :14 2 6± 32 0个 视网膜 ;(2 )Wortmannin组 (AG +IBMX +cAMP +Wortmannin) :4 14 3± 10 5 7个 视网膜 ;(3)PD980 5 9组 (AG +IBMX +cAMP +PD980 5 9) :4 15 4± 96 5个 视网膜。 结论 H89可以阻断IBMX和CPT cAMP对RGCs再生的促进作用 ,而Wortmannin和PD980 5 Objective Use three inhibitors to block three different signaling pathway to explore the mechanisms of 3-isobutyl-1-methylxanthine(IBMX) and 8 (4 Chlorophenylthio) cAMP(CPT-cAMP) on the regeneration of retinal ganglion cells. Method Fluorescent retrogratde tracing method and quantitative anatomical techniques were used to measure the numbers of RGCs in control one,control two group and experiment groups. Results 1^ In control group 1(AG),the mean number of regenerating RGC was 1428?±284/retina in 4 weeks after surgery; 2^ In control group 2(AG+IBMX+CPT-cAMP) the mean number of regenerating RGC was 4?917±1 489/retina in 4 weeks after surger; 3^ The mean number of regenerating RGC of experiment groups in 4 weeks after surgery were (1) H89 group(AG+IBMX+cAMP+H89):1?426±320/retina; (2)Wortmannin group(AG+IBMX+cAMP+Wortmannin):4143±1?057/retina; (3)PD98059 group(AG+IBMX+cAMP+PD98059):4?154±965/retina. Conclusion The promoting effects of IBMX and CPT-cAMP on regeneration of RGCs can be interrupted by H89 and can't be blocked by wortmannin and PD98059.
