摘要 目的 :以IL 6作为导向载体 ,PE4 0作为杀伤毒素 ,构建重组毒素对肿瘤细胞具有特异的杀伤功能 .方法 :通过构建IL6 (2 3 ) PE4 0重组毒素质粒 ,经大肠杆菌高效表达 ,纯化表达产物 ,进行细胞的毒性试验 .结果 :表达产物具有杀伤骨髓瘤细胞、急性髓样白血病细胞 ,胃癌细胞 ,肝癌细胞和喉癌细胞的毒性 ,当重组毒素浓度为 0 .6 μg·L-1 时可杀伤 5 0 %的SP2 / 0细胞 ,而对其他细胞无毒性 .结论 :表达的重组毒素可特异杀伤骨髓瘤细胞等 5种肿瘤细胞 ,而对试验中其他 5种肿瘤细胞及 AIM: To construct IL6 (23) PE40 recombinant toxin with IL 6 as target vector and PE40 as killer for specific killing of cancer cells. METHODS: IL6 (23) PE40 recombinant toxin was constructed and highly expressed in E. coli and the product was purified to kill cancer cells. RESULTS: The expressed product killed myeloma cells, hepatocellular carcinoma cells and promyelocytic leukemia cells. The recombinant toxin at density of 0.6 μg·L -1 could kill 50% SP2/0 cells, but not other cells. CONCLUSION: The recombinant toxin can kill 5 kinds of tumor cells including myeloma but have no effect on the other 5 kinds of tumor cells and 5 normal cells.
