摘要
目的 :观察丹参注射液对β2 糖蛋白Ⅰ (β2 glycoproteinⅠ ,β2 GPⅠ )诱导的抗磷脂综合征 (antiphospholipidsyndrome,APS)自身免疫小鼠外周血T细胞亚群和IL 2生物活性的影响 ,以探讨其对APS的治疗作用机制及免疫药理作用 .方法 :6 0只 4wk龄雌性昆明种小鼠被随机分为小、中、大剂量组及中剂量对照、模型、空白对照 6组 ,小、中、大剂量组及中剂量对照组经腹腔注射不同剂量的丹参注射液 ;1 4d后 ,小、中、大剂量组及模型组多部位皮下注射 β2 GPI检测外周血T细胞亚群和IL 2生物活性 .结果 :与模型组相比 ,小、中、大剂量组均可降低TH/TS 比值 (P <0 .0 5或P <0 .0 1 ) ;中剂量对照与空白对照组相比无统计学意义 (P >0 .0 5 ) ;药物剂量与TH/TS 比值存在负相关 (P <0 .0 1 ) .与模型组相比 ,小剂量组对IL 2活性无明显影响 (P >0 .0 5 ) ,而中、大剂量组则使之明显降低 (P <0 .0 1 ) ;中剂量对照与空白对照组无明显差异 (P >0 .0 5 ) ;药物剂量与IL 2活性存在负相关 (P <0 .0 1 ) .TH/TS 比值和IL 2生物活性呈正相关 (P <0 .0 1 ) .结论 :丹参注射液可抑制自身免疫小鼠TH/TS 比值和IL 2 ,且对空白对照组TH/TS 比值和IL
AIM: To explore the therapeutic effects and the mechanism of Chinese herbs on Antiphospholipid Syndrome (APS) by observing the effects of Salvia miltiorrhiza bunge Injection (SMBI) on anticardiolipin antibody (aCL) induced by β 2 glycoproteinⅠ(β 2 GPⅠ). METHODS: 60 female mice of 4 wk randomly fell into 6 groups. Group A was treated with small dosage, group B with mid dosage, group with C large dosage, group D with mid dosage without immunization of β 2 GPI, group E was immunized only with β 2 GPI and group F as control. Group A, B, C and D were injected through abdominal cavity with different dosages of SMBI daily and after 14 d, group A, B, C and E were immunized with 150 μg of purified human β 2 GPI in Complete Freund’s adjuvant (CFA) subcutaneously. T cell subsets were grouped by Streptavidin Biotin Complex Technique and the activity of IL 2 was measured by MTT chromatometry. RESULTS: Compared with that in group E, T H/T S ratio was reduced in group A, B and C ( P <0.05 or P <0.01) and no significant differences were found between group D and F ( P >0.05). By linear correlation, the dosage was negatively correlated with T H/T S ratio ( P <0.01). Compared with that in group E, the activity of IL 2 in group B and C decreased significantly ( P <0.01) and by linear correlation, there was a negative correlation between the dosage and the IL 2 activity ( P <0.01). There was no significant difference between group D and F ( P >0.05). There was a positive correlation between T H/T S ratio and IL 2 activity in different dilutions ( P <0.01). CONCLUSION: The suppressive effects of SMBI on aCL induced by β 2 GPI may be realized by reducing the elevated T H/T S ratio and IL 2 activity and SMBI has no effect on normal mice.
