摘要
目的 :探讨牛磺酸对仔鼠海马发育及学习记忆的影响 .方法 :大鼠自受孕开始喂以实验饲料 ,分为对照组和 6g·kg-1牛磺酸组 .测定 2 8d仔鼠海马中蛋白质、DNA和RNA的含量 ,乙酰胆碱酯酶 (AChE)活性 ,观察海马超微结构的变化及对学习记忆的影响 .结果 :仔鼠海马中蛋白质、DNA和RNA的含量 ,AChE的活性 ,6 g·kg-1牛磺酸组分别为 (7.0±3.0 ) ,(2 .5 6± 0 .13) ,(2 .7± 0 .4 )mg·g-1,(1.16± 0 .0 8)μkat·g-1;对照组为 (8.0± 2 .5 ) ,(3.2 3± 0 .5 1) ,(3.5± 0 .7)mg·g-1,(0 .99± 0 .0 7) μkat·g-1,两组差异显著(P <0 .0 1) .牛磺酸能增强学习记忆功能 .电子显微镜下对照组仔鼠海马神经元变小 ,核膜模糊不清 ,内质网数量减少 ,线粒体凝固 ,溶酶体多 ,轴突内神经丝减少 .结论 :母体在妊娠和哺乳期间缺乏牛磺酸 。
AIM: To explore the effect of taurine on hippocampus development and learning memory ability of young rats. METHODS: The rats were divided into control group and 6 g·kg -1 taurine group which had been fed with experimental feed since they were pregnant. Then the contents of protein, DNA and RNA, and the activity of AChE were determined while the change in ultrastructure of young rats hippocampus and effect on learning memory ability were observed in 28 day old young rats. RESULTS: The contents of protein, DNA and RNA, and the activity of AChE of young rats hippocampus in 6 g·kg -1 taurine group were (7.0±3.0) , (2.56±0.13), (2.7±0.4) mg·g -1 , and (1.16±0.08) μkat·g -1 , while they were (8.0±2.5), (3.23±0.51) , (3.5±0.7) mg·g -1 , and (0.99±0.07) μkat·g -1 in the control group. The difference was significant( P < 0.01). The taurine could improve learning memory ability. Electron microscopic examination revealed that, in the control group, hippocampus nerve cells were smaller, with the blurred cell nucleus membrane and indistinct, reduced endoplasmic reticulum, solidified cytochondriomes, reduced neurenteric silk in axis cylinder and increased lysosome. CONCLUSION: Lack of taurine in parent body in gestation and lactation period might affect the hippocampus development and learning memory ability of young rats.
