摘要
目的 :建立Bcl 2 /JH 基因重排的检测方法 ,调查分析Bcl 2 /JH 基因重排与非霍奇金淋巴瘤的相关性及其相关因素 .方法 :收集西安市非霍奇金淋巴瘤患者 74例的外周血单个核细胞 ,提取DNA后用PCR检测Bcl 2 /JH 基因重排 .根据Bcl 2 /JH 基因重排结果将患者分为重排组和非重排组 ,调查重排的可能危险因素 ,进行统计学处理 ;采用PCR检测了 74例非霍奇金患者血清中的HCVRNA .同时检测了献血员的HCVRNA和Bcl 2 /JH 基因重排 .结果 :74例非霍奇金淋巴瘤中有 1 8例外周血检测出Bcl 2 /JH 基因重排 ,且转位均发生在Bcl 2的主要断裂点区域 (MBR) ,HCVRNA阳性者为 6例 ,阳性率为 8.1 % .1 1 0例献血员中检测出 1例Bcl 2 /JH 基因重排 ,2例HCVRNA阳性 ,阳性率为 1 .8% .在非霍奇金淋巴瘤中Bcl 2 /JH 基因重排和HCV感染、农业暴露史有统计学关联(P <0 .0 1 ) .而与吸烟、肿瘤家族史没有关联 (P >0 .0 1 ) .结论 :Bcl 2 /JH 基因重排与非霍奇金淋巴瘤的发生具有一定的病因学联系 ;HCV感染与非霍奇金淋巴瘤Bcl 2 /JH
AIM: To develop a method to detect Bcl 2/J H gene rearrangement and to investigate the relationship between Bcl 2/J H rearrangement and non Hodgkins lymphoma and the relative factors. METHODS: Polymerase chain reaction (PCR) was used to detect Bcl 2/J H gene rearrangement with DNA extracted from blood lymphocyte and HCV RNA in 74 non Hodgkins lymphoma patients. According to the Bcl 2/J H gene rearrangement, the patients were divided into positive group and negative group. The possible risk factors of Bcl 2/J H gene rearrangement were collected and dealed with statistical method. In the mean time, we detected the HCV RNA and Bcl 2/J H gene rearrangement in donors. RESULTS: Among 74 non Hodgkins lymphoma (NHL) patients, 18 were positive for Bcl 2/J H gene rearrangement in the peripheral blood, and the rearrangement all occured in MBR, 6 were positive for HCVRNA (8.1%). Among 110 donors, there was one patient with Bcl 2/J H gene rearrangement positive, and 2 with HCV RNA positive (1.8%). Bcl 2/J H rearrangement has significant statistical relation with HCV infection and agriculture exposure ( P <0.01), However, the relationship between Bcl 2/J H rearrangement and smoking or family tumor were nonsignificant ( P >0.01). CONCLUSION: Bcl 2/J H rearrangement has some relationship with the NHLs occurrence and HCV infection.
