摘要 目的 研究转化生长因子(TGF—β1)在动脉粥样硬化形成中的作用及机制。方法 用ELISA法测定冠心病患者血TGF—β1水平,并用人动脉血管平滑肌细胞作为对象,采用MTT法、^3H—胸腺嘧啶脱氧核苷(^3H—TdR)参入法、流式细胞分析仪观察TGF—β1对流产死胎胸主动脉血管平滑肌细胞增殖的作用及机制。结果 ①冠心病组的血清TGF—β1低于正常对照组;血管病变越严重,TGF—β1水平越低。②体外培养平滑肌细胞增殖活性(MTT法、^3H—TdR参入法)随TGF—β1浓度增加而逐渐减低,且与平滑肌细胞密度无关。③随着TGF—β1浓度的升高G1期细胞比例增加、G2期细胞比例减少。结论 TGF—β1能明显抑制平滑肌细胞的增殖,人体内血TGF—β1对动脉粥样硬化形成具有保护作用。 AIM To assess the roles of trans- coronary artery disease (CHD). METHODS The forming growth f actor-beta (TGF-β1) in atherogen- cultured human vascular smooth muscle cells esis, and to investigate the changes of serum TGF- (VSMC) prepared from the human embryo (died) *1 level and its clinical significance in patients with aorta were used for experiments. Cells were exposed to different concentrations of TGF-β1 (0.1 μg · L-1 μg · L-1 and 10 μg · L-1). The proliferation of VSMC was assayed by the measurement of MTT. 3 H-TdR incorporation and flow cytome-try and human serum TGF-β1 level were measured by enzyme-linked immunosorbent assay (ELISA). RESULTS The proliferation of cultured VSMC that exposed to TGF-β1 decreased markedly, TGF-β1significantly inhibited the proliferation of cultured VSMC concentration-dependently, and this inhibitory effects was independent of SMC density. The result of flow cytometry showed that the pro- portion of cells in G1 phase increased markedly and cells in G2 phase decreased. It was observed that TGF-β1 level in patients with CHD was significantly lower than that in the controls, paticularly in those with severe vessel disease. CONCLUSION TGF-β1 can inhibit proliferation of human SMC. The TGF-β1 can protect the procession of athero-genesis.
