摘要
目的 :研究利用 NF- k B“圈套”DNA片段 ( NF- k B decoy oligodeoxynucleotides,NF- k B decoy ODN )抑制 NF- k B活性 ,对氧化压力下肝细胞脂质过氧化作用及损伤的影响。方法 :把人工合成的 NF- k B decoy ODN导入培养的鼠肝细胞 ,用 Ferric nitrilotriacetate( Fe NTA)导致鼠肝细胞经历氧化压力 ,检测培养基中 Malondialdehyde( MDA)及 L actate dehydrogenase( L DH)的量。结果 :转染 NF- k B decoy ODN的肝细胞 ,对 Fe NTA导致的肝细胞的脂质过氧化作用及损伤有显著抑制性作用 ,与对照组相比 ,MDA的值 ,前者是后者的 40 .8% ( q=10 .41,P<0 .0 1) ;L DH的值 ,前者是后者的 3 4.5 8% ( q=15 .11,P<0 .0 1)。结论 :NF- k B decoy ODN为减轻氧化压力对肝细胞的损害 。
Objective:The aim of this study was to explore the effects of NF-kB decoy ODN on cultured rat hepatocyte undergoing oxidative stress.Methods:Oxidative stress was induced by FeNTA in cultured rat hepatocytes infected NF-kB decoy ODN.The extracellular levels of MDA and LDH was evaluated.Results:NF-kB decoy ODN decreased the extracellular levels of MDA and LDH in cultured rat hepatocytes undergoing oxidative stress.Conclusion:NF-kB decoy ODN may provide a new strategy for protecting hepatocyte undergoing oxidative stress from injury and for inhibiting hepatic fibrisis.
