摘要
目的 :研究慢性肝病 ( CHD)与原发性肝癌 ( PHC)中表皮生长因子受体 ( EGFR)、基质金属蛋白酶 2( MMP2 )、基质金属蛋白酶 9( MMP9)的表达并进行比较。方法 :采用免疫组化 ( SP)法研究 47例 CHD与 PHC组织中EGFR、MMP2 、MMP9的表达。结果 :EGFR阳性定位于肝实质细胞浆中 ,MMP2 、MMP9阳性定位于肝实质细胞和间质细胞浆中。EGFR在肝癌中的表达率 ( 73 .0 8% )显著高于 CHD中 ( 2 8.5 7% ) ( P<0 .0 1)。MMP2 、MMP9在肝癌中阳性表达率完全一致 ( 80 .77% )。MMP9在 PHC中表达率 ( 80 .77% )显著高于 CHD中 ( 5 2 .3 8% ) ( P<0 .0 5 )。在 CHD中 MMP2 、MMP9表达与 EGFR有差异 ( P<0 .0 1~ 0 .0 5 )。结论 :EGFR高表达与 PHC发生有关。MMP2 、MMP9在 PHC及 CHD中作用机制可能不同。在前者能增加癌细胞浸润与转移的能力 。
Objective:The aim of this study was to investigate the expressions of Epidermal Growth Factor Receptor(EGFR),Matrix Metalloproteinase2 (MMP 2) and Matrix Metalloproteinase9 (MMP 9) in Chronic hepatocellular Disease(CHD)and Primary Hepatic Carcinoma (PHC).Methods:47 cases of CHD and PHC were studied by using immunohistochemistry staining method for monoclone antibody EGFR,MMP 2, MMP 9.Results:EGFR positive was located in hepatic parenchyma cytoplasm and MMP 2?MMP 9 positive was located in parenchyma and mesenchyma cytoplasm respectively. The EGFR expression rate 73.08% in PHC was significanly higher than that 28.57% showed in CHD(P<0.01). The expression of MMP 2 was corresponding to the MMP 9(80.77%) in PHC.The MMP 9 expression rate 80.77% in PHC was significanly higher than that 52.38% showed in CHD(P<0.05).In CHD ,there was significant difference between the expression of EGFR and MMP 2 (P<0.01),and between the expression of EGFR and MMP 9 (P<0.05) respectively.Conclusion:The higher expression of EGFR is related to the PHC tumorgenesis. There may be different machanisms of MMP 2 , MMP 9 rules in PHC and CHD .In CHD, they may decrease the liver cirrhosisgenesis and in PHC they may increase the cancer invading and metastasis.
