胰岛素对糖尿病大鼠骨代谢的影响 被引量:2

摘要 [目的 ]观察糖尿病大鼠应用胰岛素治疗后骨组织形态学改变以及血中Ⅰ型胶原氨基端肽(PINP)水平变化。 [方法 ]3月龄雄性SD大鼠 30只 ,随机分为糖尿病组 (D) ,胰岛素治疗组 (I) ,空白对照组 (C ) ,每组 10只。D组和I组大鼠经鼠尾静脉注射链尿佐菌素造成糖尿病大鼠模型 ,C组大鼠注射生理盐水。I组大鼠分别接受中效胰岛素治疗 (6~ 8u/d) ,每天两次。 9周及 14周分别处死大鼠 ,取骨与血行骨组织形态计量学分析及应用放免方法检测PINP水平。 [结果 ]9周末 ,D组和I组光镜下见骨质疏松表现 ,但 3组PINP水平无差异。 14周末D组仍见骨质疏松征象 ,而I组微观结构较 9周末明显恢复 ;D组与C组PINP水平无差异 ,I组PINP水平显著增高。 [结论 ]胰岛素可以促进糖尿病大鼠骨质的形成 ,抑制糖尿病大鼠骨质疏松的发生发展。 To determine the effect of insulin in inhibiting osteoporosis of streptozotocin-induced diabetic rats. Thirty 3-month-old male SD rats were divided randomly into three groups of 10 ,including diabetes (D), insulin treatment ( I ) and control (C) groups. Rats in group D and I were administered a single injection of streptozotocin to induce type 1 diabetes , while those in group C were physiological saline. Each rat in group I was given insulin twice a day (6-8u/d).On week 9 and 14, all rats were terminated,the lumbar spine were prepared for morphologically analysis, and the blood for detection of the aminoterminal propeptide of procollagen (PINP ) with radioimmunoassay. On week 9. Osteoporosis was found in group D and I comparing with group C, but there is no difference in the level of PINP among three groups. On week 14, there is no changes of osteoporosis in group D, but significant recovery in group I morphologically comparing with that of week 9; there is no difference between D and C, but higher in group I in the level of PINP.[Conclusion]Insulin can enhance bone formation of diabetic rats, and inhibit the development of diabetic osteoporosis.
