摘要
在浴比100的去离子水中加入0.5%的十二烷基磺酸钠(SDS)和一定量的0.05MpH7.8的磷酸缓冲液,可以98℃下快速分离各层丝胶蛋白;在聚丙烯酰胺凝胶电泳图谱上可见丝胶Ⅰ、Ⅱ、Ⅲ的不连续谱带分别为8、5、1条。初步认为,三层丝胶的蛋白质由8种、5种和1种泳动率不同的蛋白质组成,从而对各层丝胶的复合性提出了新的看法。
In the present experiment,the three layers of siricin were shown in the ratio of 48:46 :6,which is in agreement with the results of Odake Keiichi.Repeated tests showed that deionized water supplemented with 0.5% SDS(sodium dodeccyl sulphate)and certain amount of phosphoric acid as buffer liquid(pH 7.8)could separate the layers of siricin rap- idly at 98℃±and that three discontinuous bands of siricin Ⅰ,Ⅱ and Ⅲ could be distin- guished clearly in the electropherogram obtained by SDS-PAGE.Eight,five and one bands were observed in siriein Ⅰ,Ⅱ and Ⅲ,respectively.A new idea was suggested of the complexity of each siricin layer.
